I. Background | |
APLNR(APJ)是一種七次跨膜 G 蛋白偶聯(lián)受體,在人體組織中分布廣泛。Apelin 是 Apelin 受體(APLNR/APJ)的內(nèi)源性配體, Apelin/APJ 參與調(diào)控多種生理和病理過程,在 多種癌癥中 Apelin 表達(dá)增高,Apelin/APJ 通過介導(dǎo)自噬、凋亡等生物學(xué)過程調(diào)控腫瘤發(fā) 展,從而調(diào)控腫瘤細(xì)胞的增殖、遷移和侵襲。Apelin/APJ 還可以通過 PI3K-AKT、ERKMAPK 等信號(hào)通路參與免疫應(yīng)答和免疫調(diào)控。最新研究指出,APJ 與免疫檢查點(diǎn) PD-L1 存 在負(fù)調(diào)控關(guān)系。 | |
II. Introduction | |
cAMP 在 G 蛋白偶聯(lián)受體(GPCR)信號(hào)轉(zhuǎn)導(dǎo)中是一種關(guān)鍵的第二信使。當(dāng)配體結(jié)合到 GPCR,構(gòu)象發(fā)生變化,激活受體接著激活G蛋白,Gi的激活導(dǎo)致腺苷酸環(huán)化酶調(diào)控 cAMP 的下調(diào)。APLNR(APJ)藥靶模型很好的模擬了體內(nèi) APLNR(APJ)的信號(hào)轉(zhuǎn)導(dǎo)過程,原 理見下圖所示。 Figure 1. APLNR(APJ) CHO 細(xì)胞模型原理圖
