| I. Background | |
|
IL-1家族包括11種細(xì)胞因子，7種具有激動(dòng)劑活性的配體（IL-1α，IL-1β，IL-18，IL-33，IL-36α，IL-36β，IL-36γ）和4種具有拮抗活性的成員[IL-1受體拮抗劑（IL-1Rα）， IL-36Ra，IL-37，IL-38]。IL-1α，IL-1β，IL-18，IL-33，IL-36α，IL-36β，IL-36γ觸發(fā)MPKK和NF-κB導(dǎo)致炎癥反應(yīng)。而IL-1家族的其他成員具有抗炎作用。IL-1是該家族的一個(gè)標(biāo)志性細(xì)胞因子，它可以共同刺激T輔助細(xì)胞、B細(xì)胞的成熟和增殖、NK細(xì)胞的激活，并與炎癥有關(guān)。IL-1由上皮細(xì)胞、內(nèi)皮細(xì)胞、血小板、成纖維細(xì)胞和免疫細(xì)胞（如巨噬細(xì)胞和中性粒細(xì)胞）產(chǎn)生和分泌，作為先天和適應(yīng)性免疫反應(yīng)的一部分，保護(hù)機(jī)體免受損傷和感染。
IL1α或IL1β結(jié)合IL1R1誘導(dǎo)IL1R1與IL1R輔助蛋白（IL-1RAcP）二聚化，由此產(chǎn)生的復(fù)合體招募MyD88，進(jìn)而招募IL-1受體相關(guān)激酶1（IRAK1） 。IRAK1激活TNF受體相關(guān)因子6 （TRAF6）/ TGFβ活化激酶（TAK1）信號(hào)級(jí)聯(lián)，激活炎癥和免疫基因的轉(zhuǎn)錄。 |
